Zusammenfassung
Die Mechanismen der Transformation des Schultergürtels am Ursprung der terrestrischen Fortbewegung bei Wirbeltieren bleiben ein herausragendes Problem. Der Verlust von intramembranösen Knochen und die Vergrößerung von endochondralen Knochen führten zur Disartikulation des Schultergürtels vom Schädel und zur Bildung des Halses während des Übergangs von Fischen zu Tetrapoden. Trotz der funktionellen Implikationen dieses skelettalen Verschiebungsprozesses beim Aufkommen terrestrischer Wirbeltiere sind die zugrunde liegenden genetisch-entwicklungsbiologischen Veränderungen rätselhaft. Hier zeigen wir, dass sich bei Zebrafischen mesodermale Zellen des Schultergürtels, die gli3 exprimieren, einen Transkriptionsfaktor-Gen im Hedgehog-Signalweg, sowohl in intramembranöse als auch in endochondrale Knochen differenzieren. Faszinierendweise zeigten Gli- und Hedgehog-Knockout-Fische eine unerwartete Kombination aus Merkmalen aktinopterygischer Fische und Stamm-Tetrapoden-Schultergürtel. Diese ontogenetischen und anatomischen Daten deuten darauf hin, dass ein Trade-off zwischen den beiden distincten Ossifikationswegen ein tief verankertes Entwicklungsprogramm bei Knochenfischen ist und dass die Einstellung dieses Trade-offs neuartige Schultergürtel erzeugen kann, die denen von Stamm-Tetrapoden am Beginn der Wirbeltier-terrestrifizierung ähneln.

Trotz der funktionellen Implikationen dieses skelettalen Verschiebungsprozesses beim Aufkommen terrestrischer Wirbeltiere sind die zugrunde liegenden genetisch-entwicklungsbiologischen Veränderungen rätselhaft. Hier zeigen wir, dass sich bei Zebrafischen mesodermale Zellen des Schultergürtels, die gli3 exprimieren, einen Transkriptionsfaktor-Gen im Hedgehog-Signalweg, sowohl in intramembranöse als auch in endochondrale Knochen differenzieren. Faszinierendweise zeigten Gli- und Hedgehog-Knockout-Fische eine unerwartete Kombination aus Merkmalen aktinopterygischer Fische und Stamm-Tetrapoden-Schultergürtel. Diese ontogenetischen und anatomischen Daten deuten darauf hin, dass ein Trade-off zwischen den beiden distincten Ossifikationswegen ein tief verankertes Entwicklungsprogramm bei Knochenfischen ist und dass die Einstellung dieses Trade-offs neuartige Schultergürtel erzeugen kann, die denen von Stamm-Tetrapoden am Beginn der Wirbeltier-terrestrifizierung ähneln.",
